Occurrence and Distribution Patterns of Plum Tree Viruses and Genetic Diversity of Sharka Isolates in Bosnia and Herzegovina

In order to fill in a decade-long information gap regarding the biological, serological and molecular data for plum tree viruses Bosnia Herzegovina, three-phase study combining symptom evaluation, assays with high-throughput sequencing (HTS) technology was conducted. The most frequently observed symptoms were discolorations form of ring patterns, bands irregular shapes, as well vein banding. Sharka-associated patterns semicircles prevalent. A total 468 samples tested by ELISA presence PPV, ApMV, PDV, PNRSV, PBNSPaV, ACLSV MLRSV. An overall infection incidence 51.9% detected, PPV being prevalent (48.7%), followed PDV (2.99%), PNRSV (0.21%) mixed infections PPV+PDV (1.71%). RT-PCR-assisted strain typing 45 revealed PPV-D common (22.22%), PPV-REC (6.66%). Mixed PPV-D+PPV-REC detected HTS enabled recovery 9743 nts long sequence (PPV_O7/80, MW412433), which shared highest nucleotide amino acid identities isolates S13 (LC375131) from Serbia, SVN1 (LC375132) Slovenia N9 (LC375129) Bulgaria. phylogenetic analysis whole genome placed isolate D distinctive group Slovenian (LC375132). addition, (Cter)NIb/(Nter)CP fragment (MW412434) obtained this survey formed separate previously known Herzegovina (BOS64Pl BOS257Pl).
